Synergetic Protective Effects against Oxidative Stresses by Cotreatment of Centella asiatica and Follic Acid-Ferment Extracts and Phytosphingosine-1-Phoaphte in Human Dermal Fibroblasts

Abstract
The present study aims to evaluate synergetic effects of co-treatment with Centella asiatica (C. asiatica) and follic acid-ferment extracts (CFFE) and phytosphingosine-1-phoaphte (PhS1P). Cosmetic effects of each of CFFE and PhS1P are relatively well-known. However, synergetic effects of co-treatment with CFFE and PhS1P are not understood currently. Therefore, we investigated synergetic effects of co-treatment with CFFE and PhS1P and how co-treatment with CFFE and PhS1P exerts its synergetic effects. First, we determined that co-treatment with specific-concentration of CFFE and PhS1P increased cell viability. At same concentration of CFFE and PhS1P, HDFs were resisted oxidative stress-dependent cell growth arrest. In addition, we show that co-treatment of CFFE and PhS1P repressed c-Jun N-terminal kinase (JNK) activation and JNK-mediated oxidative stress response in HDFs. And co-treatment of CFFE and PhS1P-mediated blocking of JNK activation induced decrease of MMP1 mRNA expression. Therefore, co-treatment of CFFE and PhS1P synergetic protects oxidative-stress through repressing JNK phosphorylation and transcriptional activity of AP-1 complex. And these results suggest that cosmetics contained with both CFFE and PhS1P may have more therapeutic effects against oxidative-stress-mediated skin aging and formation of wrinkle.
